If you need to do this from a shell script, you could try this (where $VOLNAME is the name of your disk):
hdiutil detach `mount | sed -ne 's|^\(/dev/[^ ]*\).*/Volumes/'"$VOLNAME"'.*mounted by.*|\1|p'` -force
