You can completely control this behavior in iTunes if you set a Pause/Play trigger in Quicksilver. I use ctrl-alt-p to control this whether the iTunes window has focus or not.

Play/Pause in Quicksilver is the same as pressing the spacebar in iTunes. It depends on the symbol in the top left.
